391 Cathelicidin LL37 and skin inflammation: ex vivo study
نویسندگان
چکیده
Cathelicidin LL37 is overexpressed in plaques psoriasis. Microbial ligands are part of the triggers over-expression. [Our objectives were] to assess expression LL37, interleukins 17 and 23 skin ex vivo submitted a microbial ligand, then effects AGH8 (vegetal extract) same model. Human explants were ligand: zymosan A at 1% was applied surface on day 0, D2, D3 D6. topically after removal zymosan-disk days. Quantitative immunohistochemistry analysis LL-37, IL-17 IL-23 made (immunostaining with anti-LL37 MAB, immunofluorescence staining anti-mouse IgG. IL-17A IL23 immunostainings using anti-IL-17A or anti-IL-23 rabbit polyclonal antibodies. On D8, basal level protein control represented 18.7±7.2% epidermis surface. zymosan-exposure, LL-37 significantly increased by 228% (p<0.001) compared control. induced significant decrease 84%, untreated explants. D8 zymosan-exposed 87% (p<0.001), vs only. In this study, ligand stimulated inflammatory response human involving IL-23/IL-17 axis, Topical application zymosan-induced cascade (p<0.001).

Human cathelicidin peptide LL37 binds telomeric G-quadruplex.
Stabilization of G-quadruplex inhibits the activity of the enzyme telomerase in cancer cells. We found LL37, a host defense human cathelicidin antimicrobial peptide, to be a potent binder of G-quadruplex structures.
متن کاملStudy of tolnaftate release from fatty acids containing ointment and penetration into human skin ex vivo.
Five fatty acids (oleic, linoleic, myristic, lauric and capric) were incorporated in 10% (w/w) into ointment formulation and their influence on lipophilic model drug tolnaftate release in vitro and enhancing effect on tolnaftate penetration into epidermis and dermis of human skin ex vivo were investigated. The prepared ointments were tested for homogeneity, pH and theological properties. In vit...
متن کاملAlarmin function of cathelicidin antimicrobial peptide LL37 through IL-36γ induction in human epidermal keratinocytes.
Several dermatoses, including psoriasis, atopic dermatitis, and rosacea, alter the expression of the innate immune effector human cathelicidin antimicrobial peptide (CAMP). To elucidate the roles of aberrant CAMP in dermatoses, we performed cDNA array analysis in CAMP-stimulated human epidermal keratinocytes, the primary cells responding to innate immune stimuli and a major source of CAMP LL37 ...

The influence of cathelicidin LL37 in human anti-neutrophils cytoplasmic antibody (ANCA)-associated vasculitis
INTRODUCTION Antineutrophil cytoplasmic antibody (ANCA)-associated vasculitis (AAV) is characterised by the autoinflammation and necrosis of blood vessel walls. The renal involvement is commonly characterised by a pauci-immune crescentic glomerulonephritis (PiCGN) with a very rapid decline in renal function. Cathelicidin LL37, an endogenous antimicrobial peptide, has recently been implicated in...
